Overview of Services
The Antibody Shared Resource provides a central site for the production of antibodies crucial to the research of Cold Spring Harbor Laboratory (CSHL) Cancer Center personnel.
Antibodies are generated using either phage display technology or classical hybridoma development protocols. From ready-to-go antigen to delivered antibody takes approximately 4 months.
Monoclonal Antibodies:
CSHL Cancer Center scientists provide specific antigens, and the Shared Resource personnel handle all stages of immunization, fusion, primary screening of hybridomas, cell culture, single-cell cloning, freezing and storage of hybridoma lines, and large-scale production of monoclonal antibodies in cell culture or in vivo.
Specialized monoclonal antibody screens are coordinated with individual scientists.
The Shared Resource maintains a library of all hybridomas generated in the Shared Resource and will provide antibodies on request, pending approval of the investigator who developed them.
Phage Display:
The Shared Resource provides the reagents and expertise to carry out selection of synthetic Fabs (antibody fragments) against purified protein (0.5-1mg, no peptides).
Fabs are tagged with FLAG and can be used directly in the vast majority of applications without reformatting to full IgG format.
Since selections take place in vitro, the technology is suitable to isolate antibodies against highly homologous, non-immunogentic antigens.
